AB OBJECTIVE: To compare the serum antibody profiles of a Nigerian HIV-1
viral isolate with those of an African (Congo) and a North American
isolate (USA). METHOD: Three HIV-1 antigens, USA type IIIB, a Congolese
isolate and a Nigerian isolate (N-4), were fractionated by sodium
dodecyl sulfate-polyacrylamide gel electrophoresis and transferred to a
nitrocellulose membrane. N-4 was isolated from peripheral blood
lymphocytes from a Nigerian AIDS patient. Sera from 75 HIV-1 positive
Nigerians were reacted with each of the HIV-1 strains on Western blot.
Radioimmunoprecipitation analysis (RIPA) was performed using a positive
T cell lysate of N-4 and a negative T cell lysate (control) to
demonstrate the envelope and core specific bands. RESULTS: A majority of
sera showed reactivity to Gag and Pol gene products of all three
isolates. A few positive sera reacted with all bands, including gp120
and gp160. Reactivity to HIV-1 gp120 was confirmed by RIPA on N-4.
Cross-reactivity was observed between IIIB and some Nigerian sera.
CONCLUSION: The Nigerian isolate had relatively similar antibody
reactivity to the Congolese strain. It appears to be a prototype African
HIV-1 isolate. Further characterization of the virus and the observed
cross reactivity is ongoing at this time.
